Anions and the contraction of glycerol-extracted muscle fibers.

چکیده
Glycerinated rabbit psoas fibers 5-19 days old were immersed in contracting solutions with and without varying concentrations of different sodium salts. Raising the pH of the solution in small steps identified a threshold pH above which the fibers rapidly developed maximum tension. In solutions with added chloride or acetate (O-O.15 M), threshold pH and maximum tension changed only slightly. In solutions with added nitrate, bromide, or iodide, fibers developed much less tension, according to the series: CH&OO> Cl> NOa> Br> I-. Addition of excess calcium to the last solutions produced no further tension. After 15 min in NO,, Br-, or Isolution, fibers partially regained tension when reimmersed in acetate but not chloride solution. Fibers that had developed tension in chloride solution lost some of it when reimmersed in nitrate, bromide, or iodide solution, according to the same anion series, out they retained more tension than they could develop initially in the latter solution.

The Control of Energy Release in Extracted Muscle Fibers
Tension and P liberation were determined at the same time in glycerol-extracted muscle fibers suspended in ATP solutions. In the relaxed state, produced by ATP in rather fresh preparations, P liberation was low, but somewhat higher than in normal resting muscle. On addition of small amounts of CaCl(2) the fibers gave a strong contraction during which P liberation was on the average about 5 time...
متن کاملDifferences in the Charge Distribution of Glycerol-Extracted Muscle Fibers in Rigor, Relaxation, and Contraction
Glycerol-extracted rabbit psoas muscle fibers were impaled with KCl-filled glass microelectrodes. For fibers at rest-length, the potentials were significantly more negative in solutions producing relaxation than in solutions producing either rigor or contraction; further the potentials in the latter two cases were not significantly different. For stretched fibers, with no overlap between thick ...
متن کاملSuppression of muscle contraction by vanadate. Mechanical and ligand binding studies on glycerol-extracted rabbit fibers
The suppression of tension development by orthovanadate (Vi) was studied in mechanical experiments and by measuring the binding of radioactive Vi and nucleotides to glycerol-extracted rabbit muscle fibers. During active contractions, Vi bound to the cross-bridges and suppressed tension with an apparent second-order rate constant of 1.34 X 10(3) M-1s-1. The half-saturation concentration for tens...
متن کاملI~elaxation in Extracted Muscle Fibers* by Emil Bozler
Szent Gy~rgyi (1-3) discovered that ATP 1 produces a strong contraction in muscle fibers which have been preserved in glycerol. Later Bozler (4-7) found that such fibers, which originally are in rigor, can be transformed into a state closely resembling normal relaxation by physiological concentrations of ATP. The fibers then are very extensible and plastic, and give a strong contraction on addi...
متن کاملThe Role of Phosphocreatine and Adenosine-triphosphate in Muscular Contraction
In the presence of 20 mM PC a strong contraction is produced in glycerol-extracted muscle fibers by ATP and AMP in concentrations as low as 10(-6)M per liter. At low concentrations of nucleotide tension rises very slowly. This rise is interpreted as being due to absorption of nucleotide by the contractile elements. AMP gives an S-shaped tension curve, indicating that the conversion of AMP into ...
